Pharmacological effect | Ivermectin mainly has a good repellant effect on nematodes and surface arthropods in vivo. The mechanism of its repellent action is to promote the release of gamma-aminobutyric acid (GABA) from presynaptic neurons, thus opening GABA-mediated chloride ion channels. Ivermectin also has selectivity and high affinity for glutamate-mediated chloride ion channels located near GABA-mediated sites in invertebrate nerve and muscle cells, thus interfering with neuromuscular signal transmission, relaxing and paralysing the worm body, leading to death or expulsion of the worm body. Inhibitory interneurons and excitatory motor neurons are the sites of action in nematodes, while the sites of action in arthropods are neuromuscular junctions. The elimination rate of Hemonchodes, Osteria, Cuprea, Trichostrongylus (including Trichostrongylus Ehrsoni), Strongylus, obliquis, plectoceris, Trichocephalus, oesophagostoma, dictyocurca, and adult and fourth stage larvae of Chabot in sheep and cattle was 97% ~ 100%. It is also effective against arthropods such as maggots, mites and lice. It is slightly less effective against lice chewing and sheep tick flies. Ivermectin is also extremely effective against ticks and flies that breed in their faeces. Although the drug does not kill ticks immediately, it can affect feeding, molting and laying eggs, thereby reducing reproductive ability. The effect on blood flies is similar. The elimination rate of Ascaris, Strongyloides rubrosuis, Strongyloides lamberti, Trichocephalus, oesophagostoma, posterior strongyloides, Coryneus dentatus and immature worm bodies in pigs was 94% to 100%, and it was also very effective against trichinus spiralis in the intestine (but not trichinus spiralis in the muscles), and it also had a good control effect on blood louse and Sarcoptes suis. Not effective against flukes and tapeworms. The pharmacokinetics of ivermectin vary significantly with different animal species, dosage forms and routes of administration. The bioavailability of subcutaneous injection is higher than that of oral administration, but oral administration is more rapidly absorbed than subcutaneous injection. After absorption, it can be well distributed to most tissues, but it is not easy to enter cerebrospinal fluid. The apparent volume of distribution of cattle, sheep and pigs was 0.45 ~ 2.4L/kg, 4.6L/kg and 4L/kg, respectively. Most animals have long half-lives, 2 to 3 days for cattle, 2 to 7 days for sheep and 0.5 days for pigs. This product is metabolized in the liver, mainly hydroxylated in cattle and sheep, and mainly methylated in pigs. It is mainly excreted from feces, and less than 5% is excreted in the urine as the original form prototype or metabolite In lactating females up to 5% of the administered dose is excreted from the milk. |
